TPMT polymorphism

Intended Use

This genotyping test to intended to assessing genetic risk for severe myelosuppression with standard dosing of thiopurine drugs in individuals for whom thiopurine therapy is being considered or who have had an adverse reaction to thiopurine therapy.
Specimen Required

Lavender (EDTA), Pink (K2EDTA), FFPE

Patient Preparation
  • Sample Collection: 2 Tubes / EDTA Blood / 2 mL per tube or FFPE
  • Specimen Preparation: Do not freeze. Transport 3 mL whole blood. (Min: 1 mL)
  • Storage/Transport Temperature: Refrigerated
  • Unacceptable Conditions
    • Plasma or serum.
    • Heparinized specimens.
    • Frozen specimens in glass collection tubes.
  • Remarks: –
  • Stability: Ambient: 72 hours; Refrigerated: 1 week; Frozen: 1 month.
Methodology
The procedure is based on PCR amplification and hybridization of amplification products to a test strip containing allele-specific oligonucleotide probes. The assay covers 3 polymorphic loci in the TPMT gene.
